高效液相色谱法测定鱼用麻醉剂在鱼体中的浓度

摘    要:目的 :建立高效液相色谱 (HPLC)测定鱼血和鱼肝中麻醉剂间氨基苯甲酸乙酯甲烷磺酸盐 (MS_2 2 2 )浓度的方法。方法 :样品用重蒸水匀浆 ,离心 (12 0 0 0r/min) 15min ,将上清液用 0 .45 μm过滤膜过滤后进YWG_C18柱分析 ,流动相为 0 .2mol/L磷酸缓冲液 (pH 3.1)∶甲醇 =85∶15 ,紫外检测波长 2 2 3nm。结果 :所得标准曲线的回归方程为鱼血 :Y (ng) =0 .5 2 5 8- 0 .0 894X ,相关系数γ =0 .9939,鱼肝 :Y (ng) =- 0 .10 0 +0 .110 0X ,γ=0 .996 5 ,X为峰高。平均回收率为鱼血 87.78%、鱼肝 76 .6 6 %。结论 :本法操作简便 ,重现性和回收率均较好。

Abstract:Objective:To establish a high performance liquid chromatographic method to determine Finquel(MS-222) in fish blood and hepar.Methods:Samples were homogenized then were centrifuged (1 200r/min,15 min),the supernatant retained and microfiltered through 0.45 μm micropore membrane prior to injection onto the YWG-C- 18 ,column.The mobile phase was 0.2 mol/L phosphate buffer(pH 3.1):methanol=85∶15,the UV detection wavelength was 223 nm.Results:The calibration curve were y(ng)=0.525 8+0.089 4X,r=0.993 9 for fish blood and Y(ng)=-0.100+0.1100X,r=0.996 5 for fish hepar,X was peak height.The average recoveries were 87.78% for fish blood and 76.66% for fish hepar.Conclusions:This method is simple,rapid and reproducible,and it has a high recovery.
