| 摘 要： | 在Ｓｏｕｔｈｅｒｎ印迹法分析的基础上用ＰＣＲ方法分析了２７例小儿急性白血病细胞的免疫球蛋白重链基因ＶＤＪ区的变化。６例未分化型白血病中５例有Ｖ＿Ｈ－Ｄ＿Ｈ－Ｊ＿Ｈ基因扩增带，提示已定向Ｂ系。１３例Ｂ系来源的白血病细胞中９例发生ＶＤＪ完全重排，４例Ｂ系来源和１例未分化型白血病ＰＣＲ无阳性扩增带而Ｓｏｕｔｈｅｒｎ印迹法有重排带。１例普通型白血病ＰＣＲ有阳性扩增带而Ｓｏｕｔｈｅｒｎ印迹法呈胚系。在１４例阳性病例中有１例出现二条扩增带呈双克隆性。１例Ｔ细胞性白血病和７例非淋系白血病均为阴性与ｓｏｕｔｈｅｒｎ印迹法结果一致，对２例未分化型，２例普遍型白血病进行顺序分析，发现它们的重排格局各不相同。

| Abstract: | ased on Southern Blot ailalysis we investigated the structure of the V_H DJ_H joining regionin 27 childhood acute leukemia patients by polymerase chain reaction(PCR). The result showed:(1)The amplified bands were found in five of 6 acute undifferentiated leukemia,(2) Nine of 13B-ALL had VDJ rearrangement,the rest 4 cases were negitive in PCR but.positive in SouthernBlot.(3)One case was positive in PCR but negitive in Southern Blot.(4)Two amplified bandwere found in one of 14 positive cases of PCR, which,suggested that this might be biclonality.(5)Seven of no lymphocytic leukemia and one of T-ALL showed negitive agreed with the re-sults of Southern Blot. (6)The sequences were analyzed in the leukemic DNA from 2 of acuteundifferentiated leukemia and 2 of prepre-B acute leukemia. The data indicated that they mighthave different rearrangement patterns. |
