小檗碱对HL-60细胞增殖、凋亡及血管内皮生长因子受体2表达的影响

摘    要:本研究旨在观察小檗碱对人早幼粒白血病HL-60细胞的增殖、凋亡及血管内皮生长因子受体-2(VEGFR2)表达的影响。选用HL-60细胞体外培养,在不同浓度的小檗碱(6-96μg/ml)作用下,应用CCK-8法检测小檗碱对HL-60细胞的增殖抑制作用;应用流式细胞仪检测HL-60细胞的凋亡水平及细胞周期的分布;用RT-PCR及Western blot分别检测VEGFR2 mRNA及VEGFR2蛋白表达的变化。结果显示,小檗碱能抑制HL-60细胞的增殖,促进其凋亡,呈浓度和时间依赖关系。随着小檗碱浓度增加,G1期细胞增多,S期细胞减少,同时VEGFR2 mRNA和VEGFR2蛋白表达减少。结论:小檗碱可抑制HL-60细胞增殖,促进细胞凋亡,改变HL-60细胞周期,下调VEGFR2 mRNA及VEGFR2蛋白的表达。

Abstract:This study was aimed to investigate the effect of berberine on the proliferation and apoptosis of HL-60 cells, and the expression of vascular endothelial growth factor receptor 2 (VEGFR2) in HL-60 cells. Berberine (6 - 96 μg/ml) was added to the HL-60 cell line culture medium, the CCK-8 method was used to reveal the inhibitory effect on cell proliferation, the flow cytometry was used to determine the apoptosis rate and cell cycle in HL-60 cells treated with berberine. The expression of VEGFR2 mRNA and protein were examined by RT-PCR and Western blot respectively. The results showed that the berberine inhibited the proliferation of HL-60 cells and induced their apoptosis in dose- and time-dependent manners. With the increased concentration of berberine, the percentage of HL-60 cells in G(1) phase of cycle increased significantly, and the percantage of HL-60 cells in S phase decreased significantly. The expression of mRNA and protein of VEGFR2 decreased with the increased concentration of berberine. It is concluded that the berberine can inhibit HL-60 cell proliferation and induce HL-60 cell apoptosis. The expression of mRNA and proteins of VEGFR2 decreased after treatment with berberine.
