肾炎康复片对梗阻性肾病大鼠肾脏金属蛋白酶1组织抑制剂和细胞间黏附分子1表达的影响

摘    要:目的探讨肾炎康复片对单侧输尿管梗阻大鼠检测金属蛋白酶1组织抑制剂(tissue inhibitor of metallo-proteinase-1,TIMP-1)和细胞间黏附分子1(intercellular adhesion molecule-1,ICAM-1)的作用。方法将雄性SD大鼠60只随机分为3组:假手术组(SOR,n=20)、模型组(UUO,n=20)和肾炎康复片组(SYKFT,n=20)。从术前24小时开始,肾炎康复片组按照成人每公斤体质量2倍的剂量灌胃,余组予以同等体积生理盐水灌胃,连续14d。于术后3、7、14d分别随机处死各组大鼠,取左肾组织,Masson染色观察肾间质病理学改变,化学比色法测定肾组织匀浆中羟脯氨酸的含量,采用蛋白质印迹法TIMP-1和ICAM-1的表达情况。结果模型组肾脏呈进行性肾间质纤维化及肾间质胶原沉积的改变(P〈0.05)。肾炎康复片组在不同程度上减轻了单侧输尿管梗阻大鼠肾间质胶原的沉积和肾脏病理损害,抑制了TIMP-1的表达(P〈0.05)。但肾炎康复片对ICAM-1的作用不明显。结论肾炎康复片对单侧输尿管梗阻所致的大鼠肾脏纤维化的形成有一定的改善作用,其机制可能与抑制TIMP-1的表达有关。

Effects of Chinese herbal medicine Shenyan Kangfu Tablet on tissue inhibitor of metalloproteinase-1 and intercellular adhesion molecule-1 in rats with obstructive nephropathy

Abstract:Objective To investigate the effect of shenyan Kangfu tablet (SYKFT), a compound Chinese herbal medicine, on the expressions of TIMP-1 and ICAM-1 in rats with unilateral ureteral obstruction (UUO). Methods The rats were randomly divided into 3 groups: sham operation (SOR, n = 20), UUO (n= 20) and SYKFT group (n = 20). SYKFT group was intragastric administrated SYKFT at a dose 20 times as many adult. The other groups were intragas- tric administrated at identical voluminal normal saline. The rats were sacrificed d 3, 7, 14 respectively, after the surgery. The histological changes of renal interstitial tissues were observed by Masson staining. Total kidney collagen content was determined by measuring the amount of hydroxyproline. The expressions in protein level of tissue inhibitor of metalloproteinase (TIMP-1) and intercellular adhesion molecule-1 (ICAM-1) were assessed by Western blot analysis. Results The dynamic histological changes demonstrated that SYKFT significantly inhibited interstitial fibrosis including tubular injury as well as collagen deposition (P〈0.05). The expressions of TIMP-1 and ICAM-1 protein were significantly increased in the UUO group compared with those of SOR (P〈0.05). The up-regulations of TIMP-1 were significantly suppressed by treatment of SYKFT, whileas the down-regulation of ICAM-1 expressions were not observed. Conclusion SYKFT could inhibit renal interstitial fibrosis via suppressing the expression of TIMP-1 in rats with UUO.
