诱导型一氧化氮合酶及缺氧诱导因子-1α在人脑胶质瘤的表达意义与肿瘤血管生成关系的研究

摘    要:目的探讨诱导型一氧化氮合酶(iNOS)、缺氧诱导因子-1α(HIF-1α)在人脑胶质瘤标本表达特点及相关性,探讨HIF-1α和iNOS与肿瘤血管形成的相关性。方法收集86例人脑胶质瘤(低度恶性组42例,高度恶性组44例)和新鲜脑组织10例(对照组),应用免疫组化SP法测定HIF-1αi、NOS和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平并分析其相关性。结果免疫组化结果显示,iNOS在对照组细胞中不表达,低度恶性组胞浆中呈阳性表达,而高度恶性组呈强阳性表达,2组之间表达强度差异具有统计学意义(P<0.05);HIF-1α和VEGF在正常脑组织组和胶质瘤细胞中均有表达,其中正常脑组织均弱阳性表达,低度恶性组均呈阳性表达,而高度恶性组均呈强阳性表达,HIF-1α和VEGF在3组之间表达强度差异均具有统计学意义(P<0.05)。根据免疫组化结果进行各种抗原表达相关性分析,HIF-1αi、NOS和VEGF在低度恶性组和高度恶性组中表达均具有相关性。结论 HIF-1αi、NOS和VEGF共同参与胶质瘤细胞发生发展过程,三种基因表达强度均与胶质瘤细胞恶性度相关,恶性度越高,表达量越多;HIF-1αi、NOS和VEGF三者可能存在相互协调关系,共同促进胶质瘤生长增殖和转移过程。

Abstract:Objective To investigate the expression of hypoxia-inducible factor-1 alpha(HIF-1α) and inducible nitric oxide synthase(iNOS) in glioma,and the correlation between them and the formation of tumor vessel.Methods Total 86 cases of human brain neurogliocytoma(42 cases of low potential malignancy group,44 cases of high potential malignancy group) and 14 cases of fresh brain tissue were collected.HIF-1α,iNOS and VEGF expressions were detected by immunohistochemistry between them and the formation of tumor vessel.Results The immunohistochemistry results showed that iNOS was not expressed in the fresh brain tissue group,positive expressed in the cytoplasm of low potential malignancy group and strong expressed in the cytoplasm of high potential malignancy group.It had a statistical significance between low potential malignancy group and high potential malignancy group(P<0.05);HIF-1α and VEGF were all expressed in the cytoplasm of normal brain tissue group and neurogliocytoma group.They were all positive weakly positive expressed in the normal brain tissue group,positive expressed in the low potential malignancy group and strong expressed in the high potential malignancy group.It had a statistical significance within two groups(P<0.05);Analysis of immunohistochemistry showed that HIF-1α,iNOS and VEGF had a statistical significance not only in low potential malignancy group but also in high potential malignancy group(P<0.05).Conclusion HIF-1α,iNOS and VEGF were all participated in the tumor genesis and development.The genetic expressions of HIF-1α,iNOS and VEGF were correlated with the malignant degree of glioma.Meanwhile,HIF-1α,iNOS and VEGF maybe mediated the glioma cell proliferation and growth.
