摘    要:目的建立王不留行毛状根培养体系。方法分别用发根农杆菌R15834、R1601、R1000、A4感染王不留行叶片产生毛状根,考察不同理化因子对其生长的影响,并利用HPLC测定王不留行中黄酮苷的量。结果发根农杆菌R1601转化王不留行叶片的诱导率最高,p H值为6.0,蔗糖浓度为3%的MS培养基培养毛状根增殖速度最快,王不留行毛状根中王不留行黄酮苷量略高于种子中的量。结论王不留行成功诱导毛状根,为进一步筛选适宜的王不留行发根培养体系并调控次生代谢产物的研究奠定了基础。

Abstract:Objective To establish the culture system of Vaccaria segetalis hairy roots. Methods Agrobacterium rhizogenes R15834, R1601, R1000, and A4 were used to infect V. segetalis leaves to induce hairy roots and the influences of different physicochemical factors on its growth were investigated. The content of vaccarin was determined by HPLC. Results A. rhizogenes R1601 had the highest induction rate by converted into V. segetalis leaves, The best growth cycle of cell suspension culture was defined when cultured in liquid MS medium with pH value of 6 and sucrose concentration of 3%, vaccarin in V. segetalis hairy roots was slightly higher than that in the seed. Conclusion V. segetalis could successfully induce hairy roots, the foundation has been established for further oplimizing the proper cultural system for V. segetalis hairy roots and regulating the secondary metabolites.
