神经生长因子对小鼠腹腔巨噬细胞神经胶质细胞释放一氧化氮的影响

摘    要:目的研究神经生长因子(NGF)对正常小鼠、非肥胖性糖尿病小鼠(NOD小鼠)腹腔巨噬细胞、神经胶质细胞释放一氧化氮(NO)的影响。方法实验分对照组和不同浓度NGF实验组,NO试剂盒检测细胞培养液中NO浓度。结果①与正常小鼠相比,NOD小鼠巨噬细胞、神经胶质细胞NO释放量均明显增高(P<0.05)。②与各自对照组相比,正常小鼠、NOD小鼠各实验组巨噬细胞、神经胶质细胞NO释放量均减小。③不同浓度的NGF对小鼠巨噬细胞、神经胶质细胞释放NO的影响差异无统计学意义。结论NOD小鼠可以释放高水平的NO;NGF对正常小鼠、NOD小鼠离体培养的腹腔巨噬细胞、神经胶质细胞NO的释放有显著的抑制作用。

Abstract:Objective To investigate the effect of nerve growth factor(NGF) on release of nitric oxide(NO) from peritoneal macrophages,neuroglial cell in the normal mice and the nonobese diabetic mice(NOD).Methods The mice were allocated into control group and NGF groups with NGF at concentrations 5,10,20 ng/ml,to examine the content of NO in the fluids of cultured cell with NO kits.Result ①Compared with the normal mice,the released content of NO from peritoneal macrophages,neuroglial cell was increased significantly in the NOD mice.②Compared with that of respective control groups,the released content of NO from macrophages,neuroglial cell of the NGF groups was decreased in the normal mice and the NOD mice.③NGF of the different concentration had no difference on release of NO from the cells in the mice.Conclusion Released content of NO from the cells increase in the NOD mice,NGF can significantly inhibit release of NO from macrophages,neuroglial cell in the normal mice and the NOD mice.
