摘    要:目的:建立同时测定柴葛清热颗粒中柴胡皂苷a、d和葛根素含量的方法。方法:采用高效液相色谱法。色谱柱为Agilent C18(250mm×4.6mm,5μm),流动相为乙腈-水(梯度洗脱),流速为1.0mL·min-1,柱温为30℃,检测波长为210nm,进样量为10μL。结果:柴胡皂苷a、d和葛根素的检测浓度分别在0.014~1.120mg·mL-(1r=0.9997)、0.015~1.200mg·mL-(1r=0.9996)和0.012~0.960mg·mL-(1r=0.9998)范围内与各自峰面积积分值呈良好的线性关系;三者平均加样回收率分别为99.96%、99.80%和99.83%,RSD分别为1.67%、0.97%和1.24%(n=6)。结论:本方法简便、准确、重复性好,可用于柴葛清热颗粒的质量控制。

Abstract:OBJECTIVE: To establish the method for the content determination of saikosaponin a, d and puerarin in Chaige qin- gre granules. METHODS: HPLC method was adopted. Agilent C18(250 mm×4.6 mm, 5 μm) was used with mobile phase consisted of acetonitrile-water (gradient elution)at the flow rate of 1.0 mL.min^-1. The detection wavelength was set at 210 nm and the col- umn temperature was 30 ℃. The injection volume was 10 μL. RESULTS: The linear range was 0.014-1.120 mg.mL^-1(r=0.999 7) for saikosaponin a, 0.015-1.200 mg.mL^-1(r=0.999 6) for saikosaponin d and 0.012-0.960 mg.mL^-1(r=0.999 8) for puerarin. Average recovery rates were 99.96% (RSD= 1.67%, n=6), 99.80% (RSD=0.97%, n=6) and 99.83% (RSD= 1.24%, n=6), re- spectively. CONCLUSION: The method is simple, accurate and repeatable, and can be used for the quality control of Chaige qingre granules.
