摘    要:目的:建立穿山龙总皂苷的高效液相色谱(HPLC)指纹图谱,为其质量评价提供依据。方法:测定9批不同来源的穿山龙样品。色谱柱为Kromasil C8(250mm×4.6mm,5μm),流动相为水-乙腈(梯度洗脱),流速为1.0mL·min-1,检测波长为203nm,柱温为30℃。结果:共标示出穿山龙皂苷类化合物17个特征指纹峰;不同来源的穿山龙总皂苷色谱峰峰面积比值有一定的差异。结论:HPLC指纹图谱分析法能简便、快速地区分不同来源的穿山龙,为全面控制穿山龙药材的质量提供依据。

Abstract:OBJECTIVE: To establish the HPLC fingerprints for total saponins from Dioscorea nipponica, and to provide refer- ence for quality evaluation of it. METHODS: 9 batches of D. nipponica collected from different regions were determined. The sepa- ration was performed on Kromasil C8(250mm×4.6mm,5μm) column using water-acetonitrile (gradient elution) as mobile phase at flow rate of 1.0 mL·min^-1. The UV detection wavelength was set at 203 mn and the column temperature was 30 ℃. RESULTS: 17 peaks were identified as the characteristic fingerprints of saponins from D. nipponica and the ratio of peak area of different sam- pies were different. CONCLUSION: The method is suitable to differentiate D. nipponica from different regions conveniently, and can provide reference for quality control of D. nipponica completely.
