摘    要:目的:建立同时测定延胡索中生物碱类成分含量的方法。方法:采用高效液相色谱法。色谱柱为Phenomenex Luna-C18(250mm×4.6mm,5μm),流动相为乙腈-0.03mol·L-1乙酸铵溶液(用冰醋酸调pH值为6.0),梯度洗脱,检测波长为280nm,柱温为25℃,流速为1.0mL·min-1。结果:去氢延胡索甲素、延胡索乙素、延胡索甲素的进样量分别在0.06576~3.28800、0.10576~5.28800、0.10660~5.33000μg范围内与各自峰面积积分值呈良好线性关系(r均为0.9999);三者的平均加样回收率分别为99.0%、100.4%、98.8%,RSD分别为1.59%、1.32%、1.38(n=6)。10批延胡索商品药材中,去氢延胡索甲素、延胡索乙素、延胡索甲素的含量范围分别为0.101%~0.314%、0.032%~0.111%、0.022%~0.165%。结论:本方法准确、可靠,可作为评价延胡索质量的依据之一;且可在此方法上进一步研究延胡索的指纹图谱。

Abstract:OBJECTIVE: To establish the method for the content determination of 3 kinds of alkaloids in Corydalis yanhusuo. METHODS: HPLC method was adopted. The determination was performed on Phenomenex Luna-C18(250 mm×4.6 mm, 5μm) col- umn with mobile phase consisted of acetonitrile-0.03 mol. L^-1 ammonium acetate (pH value of glacial acetic acid adjusted to 6.0, gradient elution) at the flow rate of 1.0 mL.min^-1. The detection wavelength was set at 280 nm and column temperature was 25℃. RESULTS: The linear ranges were 0.065 76-3.288 00 μg for dehydrocorydaline, 0.105 76-5.288 00μg for tetrahydropalmatine, 0.106 60-5.330 00μg for corydaline (r=0.999 9). Average recoveries were 99.0%, 100.4%, 98.8%, respectively. RSDs were 1.59%, 1.32%, 1.38% (n=6). In 10 batches of C yanhusuo, the contents of dehydrocorydaline, tetrahydropalmatine and coryda- line were 0.101% -0.314% , 0.032%-0 .111% and 0.022% -0.165%. CONCLUSION: The method is accurate and reliable, which can be used as one of the methods to evaluate the quality of C yanhusuo. On the basis of it, the fingerprints of C yanhusuo can be further studied.
