龙胆草水提物对细菌性、霉菌性阴道炎模型大鼠的保护作用研究

摘    要:目的:研究龙胆草水提物对细菌性、霉菌性阴道炎模型大鼠的保护作用。方法:往大鼠阴道注入大肠杆菌-金黄色葡萄球菌混合菌液复制细菌性阴道炎模型。实验分为空白对照(等容生理盐水)、模型(等容生理盐水)、氧氟沙星(0.06g·kg-1)和龙胆草水提物高、中、低剂量(20、10、5g·kg-1)组,灌胃给药,每天3次,连续给药7d后取大鼠阴道分泌物涂片镜检,以评定总有效率。摘除大鼠双侧卵巢,皮下注射乙烯雌酚2mg·kg-1诱发大鼠假动情期,往大鼠阴道注入白色念珠菌液复制霉菌性阴道炎模型。实验分为空白对照(等容生理盐水)、模型(等容生理盐水)、特比萘芬(0.022g·kg-1)和龙胆草水提物高、中、低剂量(20、10、5g·kg-1)组,灌胃给药,每天3次,连续7d后取阴道分泌物进行念珠菌培养,计算转阴率。结果:龙胆草水提物高、中剂量组治疗细菌性阴道炎有效率分别为100%、60%,霉菌性阴道炎转阴率分别为80%、40%,与模型组比较均有显著性差异(P<0.01或P<0.05)。结论:龙胆草水提物对大鼠细菌性、霉菌性阴道炎有一定的改善作用,其机制可能与其直接抑菌有关。

Protective Effect of Gentian Extract against Bacterial and Mycotic Vaginitis Model Rats

Abstract:OBJECTIVE: To study the protective effect of gentian aqueous extract on bacterial and mycotic vaginitis model rats. METHODS: Bacterial vaginitis model was induced by vaginal implantation of Escherichia coli and Staphylococcus aureus mixture. Those rats were divided into blank control group (constant volume of normal saline), model group (constant volume of normal sa- line), ofloxacin group (0.06 g.kg^-1), gentian extract high-dose, medium-dose and low-dose groups(20, 10,5 g.kg^-1). They were given medicine intragastrically 3 times a day for consecutive 7 days, and the total effective rate was evaluated by rat vaginal smear microscopic examination. After bilateral ovariotomy of rats, false estrus of rats was induced by subcutaneous injection of diethylstil- bestrol 2 mg.kg^-1, and then vaginal implantation of Candida albicans liquid induced mycotic vaginitis in rats. Those rats were divid- ed into blank control group (constant volume of normal saline), model group (constant volume of normal saline), terbinafine group (0.022 g. kg^-1), gentian extract high-dose, medium-dose and low-dose groups (20, 10,5 g. kg^-2). They were given medicine intragastrically 3 times a day for consecutive 7 days. Then vaginal discharge was collected for Candida culture to calculate negative conversion rate. RESULTS: The effective rates of high-dose and medium-dose gentian aqueous extract on bacterial vaginitis were 100% and 60% ; negative conversion rate of mycotic vaginitis in gentian extract high-dose, medium-dose groups were 80% and 40%, there was significantly different from that of model group(P〈0.01 or P〈0.05). CONCLUSION: Gentian aqueous extract can relive bacterial and mycotic vaginitis in rats, and its mechanism may be related directly with its bacteriostasis.
