松潘乌头总碱的抗炎机制研究

摘    要:目的:研究松潘乌头总碱(TAS)的抗炎机制。方法:采用不同致炎剂复制大鼠、小鼠炎症模型,检测其毛细血管通透性、多形核白细胞百分比和炎症组织前列腺素E(PGE)、丙二醛(MDA)含量及血清超氧化物歧化酶(SOD)活性。结果:3.0、1.5mg.kg-1TAS对乙酸致小鼠腹腔毛细血管通透性的增加与明胶诱发的多形核白细胞游走均有明显抑制作用;2.0、1.0mg.kg-1TAS可使角叉菜胶致炎大鼠炎症组织中PGE、气囊灌洗液中MDA含量降低,同时提高血清SOD活性。结论:TAS的抗炎机制与其降低毛细血管通透性、抑制多形核白细胞游走与抑制炎症介质PGE的释放和清除自由基、抑制脂质过氧化有关。

Abstract:OBJICTIVE: To study anti-inflammatory mechanism of total alkaloids of Aconitum sungpanense (TAS). METHODS:The inflammatory models were induced by different inflammatory agents in mice and rats. The vascular permeability, leucocyte migratory response and the content of PGE and MDA in inflammatory tissue, and the activity of SOD in serum were measured. RESULTS: TAS (3.0, 1.5 mg.kg-1) inhibited the increase of vascular permeability induced by HAC and the leucocyte migratory response caused by gelatin in mice. TAS (2.0,1.0 mg. kg 1) also decreased the contents of PGE and MDA in inflammatory tissue induced by carrageenin, and increased the activity of SOD in serum in rats. CONCLUSION: The anti-inflammatory mechanism of TAS are related to the reduction of vascular permeability, leucocyte migratory response and PGE release, the clearing of free radicals and inhibition of lipid peroxidation.
