褐藻多糖硫酸酯精提物对慢性肾衰模型大鼠的保护作用研究

摘    要:目的:研究褐藻多糖硫酸酯精提物(RFSE)对慢性肾衰模型大鼠的保护作用。方法:70只大鼠随机分为正常对照(1%羧甲基纤维素钠)、模型(1%羧甲基纤维素钠)、地塞米松(0.1mg.kg-1)、褐藻多糖硫酸酯(FS,100mg.kg-1)和RFSE高、中、低剂量(100、50、25mg.kg-1)组。灌胃365mg.kg-1腺嘌呤(每天1次,连续20d)以复制慢性肾衰模型。模型复制成功后,除正常对照组外,各组均灌胃给药,每天2次,连续25d,末次给药后测定24h尿量、尿蛋白、红细胞计数(RBC)、血红蛋白(Hb)、肌酐(Scr)、尿素氮(BUN)、Ca2+和P3+,并行肾脏组织形态学观察。结果:高、中、低剂量RFSE均能减少模型大鼠24h尿量、尿蛋白的排泄,降低血清Scr、BUN和P3+的水平(P<0.01或P<0.05);高、中剂量RFSE还可减轻肾衰模型大鼠的肾脏病理改变(P<0.01)。结论:RFSE对腺嘌呤所致的大鼠慢性肾衰有明显的保护作用。

Abstract:OBJECTIVE: To investigate the protective effect of refined fucoidan sulfate extraction (RFSE) on chronic renal failure (CRF) in rats. METHODS: 70 SD rats were randomly divided into 7 groups, i.e. normal control group (1% sodium carboxymethylceUulose), model group (1% sodium carboxymethylcellulose), dexamethasone group (0.1 mg.kg^-1), fucoidan sulfate (FS, 100 mg.kg^-1) group and RFSE high, middle and low-dose groups (100, 50, 25 mg.kg^-1).CRF model was built successfully by adenine 365 mg.kg^-1 intragastrically once a day for consecutive 20 day. Those groups were feed drugs for successive 25 days twice a day intragastrically. The 24 h urine volume, urine protein, RBC, Hb, Set, BUN, Ca2+ and p3+ were determined after the last administration, Histomorphology of kidney was observed. RESULTS: It was observed that urine volume and protein in 24 hours, and the levels of Scr, BUN, and p3+ in high, middle, and low-dose of RFSE groups were significantly lowered (P〈0.01 or P〈0.05). Moreover, the pathological changes were improved in the high and medium doses of RESE groups(P〈0.01). CONCLUSION: RFSE has an obviously protective effect on the adenine induced CRF in rats.
