根尖周病变中破骨细胞分化因子的免疫组化定位

摘    要:目的:观察大鼠根尖周病变过程中破骨细胞分化因子(osteoclast differentiation factor, ODF)的表达,探讨ODF与破骨细胞及根尖周骨吸收的关系。方法:将25只 SD大鼠开髓,分别于术后 0,7,14,21,28 d取下颌骨,制备组织切片,分别用免疫组织化学和酶组织化学的方法检测 ODF及破骨细胞在根尖周病损中的表达与分布。结果:在正常根尖周组织中偶见 ODF表达阳性细胞和前破骨细胞;术后 7 d根尖周有炎症细胞浸润,可见较多的ODF阳性细胞和少量破骨细胞;术后14 d根尖周表现为急性炎症反应,ODF阳性细胞及破骨细胞的数量上升至高峰;术后21~28 d根尖周病损进入慢性炎症期,ODF表达迅速下降,破骨细胞数量明显减少。结论:ODF表达于根尖周病损中,可能诱导了破骨细胞的分化,导致牙槽骨的吸收。

Abstract:Objective: To observe the kinetics of osteoclast differentiation factor(ODF) and osteoclast expression in the experimental periapical lesions in rats. Methods: Twenty-five SD rats were sacrificed at day 0, 7, 14, 21 and 28 respectively after the mandibular first molar pulps were exposed. The jaws that contained the first molar were obtained and tissue slices were routinely prepared. Immunocytohistochemistry and enzymehistochemistry techniques were applied to detect the expression of ODF and osteoclasts in the periapical lesions. Results: The expression of ODF and osteoclasts were observed at day 7. The expression and number of RANKL-positive cells and osteoclasts were climaxed at day 14. In the same period, inflammatory cell infiltration and alveolar bone resorption were observed in the periapical lesions and deteriorated. In the 21st and 28th-day-samples, the expression of ODF decreased and fewer osteoclasts could be observed. Conclusion: ODF was observed in the periapical lesions. This protein may be involved in the differentiation of preosteoclasts into osteoclasts and then be responsible to the following periapical alveolar bone resorption.
