参芎注射液对大鼠脑缺血再灌注损伤的保护作用

摘    要:目的观察参芎注射液对脑缺血再灌注大鼠行为学、脑梗死体积和组织病理学的影响,探讨其脑保护作用及机制。方法33只雄性SD大鼠随机分为4组假手术组、对照组、川芎嗪组和参芎组。采用线栓法制作大脑中动脉缺血1h再灌注模型,用Longa5分制评分和氯化三苯四氮唑(TTC)染色法评价行为学和脑梗死体积,再灌注24h时电镜观察缺血边缘区超微结构改变。结果各组在再灌注6h和24h时,Longa评分无显著差异,但参芎组再灌注24h时的Longa评分较再灌注6h时显著改善(P<0·05);川芎嗪组和参芎组的梗死体积较对照组显著缩小(P<0·05),川芎嗪组和参芎组两组比较无显著差异;川芎嗪组、参芎组细胞的损伤程度较对照组显著减轻(P<0·05);在电镜下,参芎组缺血边缘区的神经细胞超微结构改变轻于对照组,优于川芎嗪组。结论参芎注射液对脑缺血再灌注损伤有一定的保护作用,且优于单用川芎嗪。

Abstract:Objectives: To observe the effect of Shenxiong injection (salivae miltiorrhizae, liguspyragine hydrochloride and glucose injection) on behavioral changes, infarct volume and histopathology changes after cerebral ischemia/reperfusion in rats, and to explore its neuroprotective effect and mechanism. Methods: Thirty three male SD rats were randomly assigned to four groups: sham-operated group, control group, ligustrazine-treated group and Shenxiong-treated group. Cerebral ischemia (1 hour)/reperfusion model was established by occluding middle cerebral artery with intraluminal suture method. The behavioral changes and cerebral infarct volume were evaluated by the Longa's score (5 points) and triphenyltetrazolium chloride (TTC) staining, and the ultrastructure changes in the ischemic margin were observed under the electron microscope at 24 hours after reperfusion. Results: There were no significant differences among the 4 groups on Longa's score both at 6 and 24 hours after the reperfusion. However, the Longa's score in the Shenxiong-treated group was improved more significantly at 24 hours after the reperfusion than that at 6 hours after the reperfusion (P<0.05). Compared with the control group, the cerebral infarct volume in the Shenxiong-treated group and ligustrazine-treated group decreased significantly (P<0.05), but there was no significant differences between Shenxiong-treated group and ligustrazine-treated group. The severity of neuron damage both in Shenxiong-treated group and ligustrazine-treated group was reduced more significant than that in the control group (P<0.05); the neuron changes of ultrastructure in the ischemic margin were lighter in the Shenxiong-treated group than those in the control group under the electron microscope, and it was better than the ligustrazine-treated group. Conclusions: Shenxiong injection may have certain neuroprotective effects on cerebral ischemia/reperfusion injury, which is better than ligustrazine alone.
