Abstract:Objective To investigate the effects of 7-difluoromethyl-5,4’-di-n-octylgenistein(DFOG) on the growth of human lung caner A549 cells and its mechanism.Methods A549 cells were cultured in vitro and were treated with various concentrations of DFOG.The ability of cell colony formation was detected by the colony formation on plate.The cell cycle phase distribution was determined by flow cytometry(FCM) analysis.The expression of FoxM1 was analyzed by Western blot.Results FCM analysis showed that DFOG resulted in cell cycle arrest in G2 phase.The clonogenic assay indicated that DFOG significantly inhibited the anchorage-dependent growth of A549 cells(P<0.05).Exposure of A549 cells to DFOG(2.0 μmol/L) at 6 h,12 h and 24 h,the level of FoxM1 expression decreased,in time-depentment manner.Conclusion DFOG inhibited the growth of A549 cells is associated with downregulation of FoxM1 expression.
