白血病细胞雄激素受体基因启动子甲基化状态的研究

摘    要:目的 检测白血病细胞雄激素受体基因启动子区CpG岛甲基化状态情况,初步探讨雄激素受体基因启动子区甲基化与白血病发病机制的可能联系.方法 应用甲基化特异性PCR技术(methylation specific PCR, MSP)检测3种白血病细胞株及15例患者骨髓标本雄激素受体基因甲基化状态.结果 白血病细胞株均出现甲基化和非甲基化带,呈部分甲基化状态,而作为对照的正常人白细胞均只出现非甲基化带.15例白血病患者全部检出甲基化状态. 结论 白血病细胞的雄激素受体基因启动子区存在高甲基化现象,可能为白血病的致病机制分析提供了一个新的方向,提示AR基因启动子CPG岛的甲基化有可能成为白血病的新的参考标记物.

Abstract:Objective To investigate the methylation status of CpG islands of the androgen receptor (AR) gene promoter in leukemic cells, and try to find the possible relationship between AR methylation and pathogenic mechanism of leukemia. Methods The methylation status of CpG island of AR gene promoter in 3 leukemic cell lines and 15 leukemic bone marrow cells was detected by methylation specific PCR(MSP). Results The cell lines showed both methylation bands and unmethylation bands, whereas normal white blood cells only showed unmethylation bands. The methylation band of AR promoter was observed in 15 leukemic bone marrow cells. Conclusion The CpG islands of AR promoters are hypermethylated in 3 leukemia lines and leukemic bone marrow samples researched. This study may provide a new direction to study the pathogenic mechanism of leukemia, and indicates that AR promoter methylation could be a potential reference marker for leukemia diagnosis.
