| Abstract: | Objective To investigate the effects of Dongchongxiacao on human mesangial cells （ HMCs ） cultured in vitro and explore the nephroprotective mechanism of Dongchongxiacao for diabetic nephropathy. Methods We get the rats serum with different concentration of medicine, and then study the effects of the serum on HMCs. the HMCs proliferation was assessed by MTT assay. RT-PCR was used to detect tile expression of transforming growth factor beta lmRNA （ TGF- β1mRNA ）. Results The high-dose serum can inhibit the proliferation of mesangial cells under high glucose condition at 24h,g8h （ P 〈 0.01 ）, The expression of TGF- β1mRNAof the high-dose are higher than control group （ P 〈 0.05 ）. Conclusion Dongchongxiacao not only can inhibit high glucose-induced proliferation of HMCs, but also can decrease the secretion of TGF- β1, so to delay the progression of DN. |
