摘    要:目的:探讨多抗甲素(PA)能否增强LAK细胞增殖反应及LAK活性;方法:采用MTT方法测定小鼠脾淋细胞的增殖反应及LAK细胞杀伤S_(180)肉瘤及K_(562)人红白血病细胞的作用;结果:①亚适剂量IL—2(200U/ ml)和不同浓度PA共同诱导脾淋巴细胞,PA能明显增强淋巴细胞的增殖反应且呈剂量依赖性;②在较高浓度时,PA能明显增强LAK细胞杀伤S_(180)细胞作用,对于LAK细胞杀伤K_(562)细胞,无论浓度高低,PA均有明显增强作用;结论:在临床上可望将PA与LAK/IL—2联合应用,以提高其抗瘤效果,降低IL—2用量及毒副作用。

EFFECTS OF POLYACTIN A ON PROLIFERATION AND CYTOTOXICITY OF MOUSE SPLEEN LAK CELLS
Abstract:In order to study the effects of polyactin A(PA), a novel responsive modifier, on LAK activities, spleen LAK proliferation and cytotoxicities of mouse for S180 and K562 tumor cell lines were determined with MTT colorimetric assay, The results showed that when mouse spleen cells were cultivated in the presence of PA and interleukin 2(IL-2), the cell's proliferation was enhanced significantly in a dose-dependent manner. PA could remarkably increase LAK cytotoxicity for S180 rumor cell in higher concentration, however, it could increase LAK cytotoxicity for K562 tumor cell whether its concentration was low or high. It is suggested that if PA be used to treat tumor together with LAK/IL -2 biothera-py, the treatmental efficacy would be raised and IL - 2 dosage and its side-effects would reduce.
