肝炎平对家兔血吸虫肝纤维化MMP-2、 TIMP-1表达的影响

目的　研究肝炎平抗家兔血吸虫肝纤维化的作用, 探讨其抗肝纤维化的可能机制。方法　实验分为3 组:正常对照组(N组)、血吸虫感染 吡喹酮组(A组) 及血吸虫感染 吡喹酮 肝炎平治疗组(B组)。采用苏木精伊红染色及Masson染色, 观察肝组织病理学改变; 采用免疫组织化学SP 法检测各组肝组织基质金属蛋白酶2(MMP-2)、金属蛋白酶组织抑制因子- 1 (TIMP- 1) 的表达水平。结果　B组肝组织胶原纤维含量低于A组(P<0 05); B组MMP 2、TIMP -1的表达水平均低于A组(P<0 .01, P<0 .05)。结论　在抗血吸虫治疗后使用肝炎平,能通过减少肝组织MMP- 2、TIMP- 1的表达而发挥抗血吸虫肝纤维化的作用。

Effect of Ganyanping on the Expression of MMP-2 and TIMP-1 in Rabbits with Schistosomal Liver Fibrosis

Objective To study the antifibrotic effect and possible mechanism of Ganyanping on schistosomal liver fibrosis of rabbits after anthelmintic treatment.Methods The rabbits were randomly divided into 3 groups: normal control group (N), schistosomal infection praziquantel group (A) and schistosomal infection praziquantel Ganyanping group (B). The histopathological changes of liver were observed by HE and Masson staining. The expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in liver tissue was detected by immunochemistry.Results The contents of collagenous fibers in group B were lower than in group A (P<0.05). The expression of MMP-2 and TIMP-1 was decreased in group B as compared to group A (P<0.01,P<0.05).Conclusion Ganyanping,after anthelmintic treatment,could reduce the expression of MMP-2 and TIMP-1 in liver of schistosomal liver fibrosis,which may be one of the antifibrotic mechanisms of Ganyanping.
