益气类中药对肺纤维化大鼠模型肺组织中TNF-α、IL-8的影响

目的：观察博莱霉素所致肺纤维化模型大鼠肺组织中TNF-α和IL-8的表达水平,探讨益气类中药对大鼠肺纤维化的作用及其机制。方法：32只SD大鼠随机分为空白组、模型组、益气组和泼尼松组,采用气管内滴加博莱霉素的方法制作肺纤维化模型,并应用相应的药物灌胃干预,各组大鼠分别于第28 d处死,观察大鼠肺组织匀浆中TNF-α、IL-8的表达水平。结果：与空白组比较,模型组、泼尼松组大鼠肺组织中TNF-α、IL-8的表达水平明显增高（P〈0.01）,益气组IL-8的表达水平明显增高（P〈0.01）、TNF-α表达水平无明显差异（P〉0.05）。与模型组比较,益气组和泼尼松组肺纤维化程度明显减轻（P〈0.01）,肺组织中TNF-α、IL-8表达水平也显著降低（P〈0.01）。益气组较泼尼松组肺纤维化程度更轻（P〈0.05）,肺组织中TNF-α、IL-8表达水平也更低（P〈0.01）。结论：益气类中药与泼尼松均可下调肺组织中TNF-α及IL-8的表达水平,抑制或延缓肺纤维化的发生发展,益气类中药疗效较泼尼松更好。

Effect of Yiqi of Traditional Chinese Medicine on TNF-in Lung Tissue of Pulmonary Fibrosis in the Rat Model of Alpha,IL-8

Objective： To observe the expression level of TNF- alpha and IL- 8 in lung tissue of pulmonary fibrosis induced by bleomycin model in rats,to investigate the effect of Yiqi of traditional Chinese medicine on pulmonary fibrosis in rats and its mechanism. Methods： 32 SD rats were randomly divided into blank group,model group,Yiqi group and prednisone group,making the pulmonary fibrosis model adopts the method of intratracheal bleomycin plus,and apply the appropriate drug stomach intervention,the rats were sacrificed at twenty- eighth D,observe the expression level of TNF-alpha,IL- 8 uniform slurry in the lung tissues of rats. Results： compared with the blank group,the model group,the expression level of TNF- alpha,pour IL- 8 in lung tissue of rats in prednisone group were significantly increased（ P〈0. 01）,the expression level of IL- 8 in the Qi group increased significantly（P〈0. 01）,no significantdifferences in the level of expression of TNF-（P〈0. 05）. Compared with the model group,Yiqi group and prednisone group,pulmonary fibrosis significantly reduced（P〈0. 01）,lung tissue TNF- alpha,IL- 8 expression was significantly decreased（ P〈0. 01）. Qi group than the prednisone group,pulmonary fibrosis and lighter（ P〈0. 05）,lung tissue TNF- alpha,IL- 8expression is lower（P〈0. 01）. Conclusion： the expression level of TNF- alpha and IL- 8 Qi and Chinese medicine and prednisone can be down regulated in lung tissue,inhibit or delay the occurrence and development of pulmonary fibrosis,Qi and Chinese medicine curative effect better than prednisone.
