酸苦克甘方对糖尿病大鼠胰岛β细胞形态、功能影响

摘    要:目的:评价临床常用经验用中药方剂对2型糖尿病大鼠模型胰岛β细胞形态、功能影响,为其临床应用提供实验依据。方法:采用随机、对照试验方法将小剂量链脲佐菌素诱导建立的T2DM动物模型随机分成3组:生理盐水对照组、中药汤剂治疗组(分常规剂量组与高剂量组),治疗4周,测治疗前后空腹血糖,空腹血清胰岛素、C肽,计算胰岛素敏感指数(ISI)。治疗后光镜下观察胰岛细胞组织。结果:治疗4周后治疗组空腹血糖、血清胰岛素均有有不同程度下降,其中常规组较高剂组下降明显,具有统计学意义。治疗组C肽均有升高,其中常规剂量组上升较明显。与盐水模型组相比,常规组及高剂组ISI呈上升趋势。与对照组相比,两组治疗组胰腺组织胰岛细胞数量及炎症反应均有不同程度的改善。结论:酸苦克甘方能降低链脲佐菌素诱导建立的T2DM动物模型的空腹血糖及空腹血清胰岛素水平,并能增加C肽值,增加胰岛素敏感性,改善胰岛功能。使胰岛细胞数量明显增加,炎性细胞明显减少,可以促进胰岛β细胞再生和修复,延缓或阻止胰岛β细胞的衰竭。

Abstract:Objective:make an attempt to evaluate The influence of the form and function of Acid bitter against Gan Fang on type 2 diabetic rats islet beta cells,then provide the experiment evidence for clinical application. Methods:Random and controlled trial method was adopted in this experiment. 60 type 2 diabetic rats induced by small dose STZ were randomly divided into three groups: aline therapy control group and herb tea therapy group( divided into common dose group and high dose group),Levels of FBG、FINS and C- P of the three groups were recorded before the beginning of the treatment and at the end of the 4treatment weeks; ISI was calculated also. Islet beta cells were observed under light microscope after the treatment. Results:All three groups were able to decrease FBG and FINS to a certain levels after 4 weeks therapy. Among them the common dose therapy group decreased more significantly(P〈0. 05). C- P were increased both in the herb tea therapy groups,but the common dose therapy group increased more significantly( P〈0. 05). Compared with aline therapy control group,ISI shows an increasing trend in both two therapy groups,inflammation and the number of pancreas island βcells could also be ameliorated. Conclusions:Acid bitter against Gan Fang could effectively depress the FPG and FINS,increase C- P and ISI,ameliorate founction of pancreas island βcells. It also can increase the number of pancreas island βcells significantly,reduce inflammation,promote regrneration and repair,delay or prevent failure of βcells.
