补虚消癥汤对大鼠肾小球系膜细胞MMPs及TIMPs表达的影响

目的：观察补虚消癥汤对大鼠肾小球系膜细胞基质金属蛋白酶（Matrix metalloproteinase,MMPs）及基质金属蛋白酶组织抑制剂（Tissue Inhibitor of metalloproteinase,TIMPs）表达的影响,探讨补虚消癥汤对肾小球硬化的防治作用。方法：（1）应用血清药理学方法,制取中药的药理血清。（2）采用肿瘤坏死因子-α（Tumor Necrosis Factor,TNF-α）作为刺激因子,诱导大鼠肾小球系膜细胞,使其发生增殖。（3）应用实时荧光定量PCR法、酶联免疫吸附ELISA法观察各组间MMP-2、MMP-9、TIMP-1及TIMP-2mRNA表达的情况。结果：1）补虚消癥汤能有效提升MMP-2/TIMP-2的比值,与对照组比较差异,高、中浓度组有统计学意义（P〈0.05）;2）补虚消癥汤能有效提升MMP-9/TIMP-1的比值,与对照组比较差异,均有统计学意义（P〈0.05）。结论：补虚消癥汤能通过影响MMP-2、MMP-9、TIMP-1及TIMP-2等的表达,改善MMP-2/TIMP-2、MMP-9/TIMP-1平衡,从而使细胞外基膜降解增加,缓解肾小球损伤。

Influence of Buxu Xiaozheng Decoction on Mesangial Cells MMP and TIMP Expressions

Objective： To study the effect of Buxu Xiaozheng Decoction on rat mesangial cell Matrix metalloproteinase（MMPs） and tissue inhibitor of metalloproteinase（TIMPs） expressions,and to explore Buxu Xiaozheng Decoction＇s effect on glomerulosclerosis. Methods：（1）Using serum pharmacological method,preparation of traditional Chinese medicine pharmacological serum was carried out.（2） Tumor Necrosis Factor（TNF- α） as a stimulating factor,rat mesangial cells were to proliferate.（3） PCR and ELISA were used to observe the groups; MMP- 2,MMP- 9,TIMP- 1 and TIMP- 2mRNA expressions. Results：（1） Buxu Xiaozheng Decoction can effectively enhance the MMP- 2 /TIMP- 2 ratio and the difference compared with control group,high,medium concentration group was statistically significant（ P〈0. 05）.（2） Buxu Xiaozheng Decoction can effectively improve MMP-9/TIMP-1 ratio and compared with the control group differences were statistically significant（P〈0. 05）. Conclusion： Buxu Xiaozheng Decoction through impacting MMP- 2,MMP- 9,TIMP- 1 and TIMP- 2 expressions to improve the MMP- 2 /TIMP- 2,MMP- 9 /TIMP- 1 balance,so that cells increased degradation of extracellular matrix film and alleviate glomerular injury.
