当归注射液对缺血/再灌注损伤大鼠心肌P57~(kip2)和CDK2蛋白表达的影响

摘    要:目的:观察当归注射液对缺血/再灌注损伤(IRI)心肌细胞P57kip2和CDK2蛋白表达的影响。方法:30只SD大鼠构建心肌IRI模型后随机分成假手术组、缺血/再灌注损伤(IRI)组和当归组,免疫组化法观察各组心肌细胞内P57kip2和CDK2蛋白表达情况,测定其平均光密度和阳性面积,并与正常对照组比较。结果:IRI组心肌细胞内P57kip2蛋白表达的平均光密度和阳性面积率均低于正常对照组和假手术组(P<0.01),而CDK2蛋白表达较高(P<0.01);当归组P57kip2和CDK2蛋白表达与正常对照组和假手术组无差异(P>0 05)。结论:当归注射液对大鼠缺血/再灌注损伤心肌细胞有保护作用。

Abstract:Objective To observe the effect of angelica injection on the expression of P57kip2 and CDK2 in rats with myocardial ischemia-reperfusion injury.Methods Myocardial ischemia-reperfusion injury(IRI)model were prepared in 30 SD rats,and another 5 rats were selected as normal control.Then the rats were randomly divided into sham-operation group,IRI group and angelica treated group.The expression of P57kip2 and CDK2 in myocardium were examined with immunohistochemical technique.The mean optical density and positive area were determined and compared with control group.Results The mean optical density and positive area of myocardial P57kip2expression in IRI group were obviously lower than those of control group and sham-operation group(P0.01),on the contrary,the expression of CDK2 were significantly higher in IRI group(P0.01);the expression of P57kip2 and CDK2 in angelica group had no significant difference with control group and sham-operation group(P 0 05).Conclusion Angelica injection may play an important role in protecting myocardium from ischemia reperfusion injury.
