| 摘 要： | 目的探讨抗人胰腺癌单克隆抗体SZ-121的制备并对其进行初步鉴定。方法用人胰腺癌细胞株Patu8988作为免疫原,利用杂交瘤技术制备抗人胰腺癌单克隆抗体,应用流式细胞术和免疫组化鉴定抗体的特异性。结果成功获得一株能够稳定分泌特异性抗体的杂交瘤细胞系SZ-121,该抗体与胰腺癌组织呈阳性反应,与胰腺良性病变组织呈弱阳性反应。结论单抗SZ-121对胰腺癌的早期诊断可能具有一定的潜在价值。

| Abstract: | Objective To prepare and identify monoclonal antibody（McAb） of human pancreatic cancer associated antigen.Methods The monoclonal antibody against human pancreatic cancer was produced using hybridoma technique with Patu8988 cell as immunogen,and its specificity was identified with immunochemical SP staining and immunohistochemistry.Results A monoclonal antibody was obtained successfully,and named SZ-121.The monoclonal antibody SZ-121 had positive reaction with tissue of human pancreatic cancer,and low positive reaction with the tissue of pancreatic benign lesion.Conclusion SZ-121 may be useful for the diagnosis of pancreatic cancer in the early stage. |
