摘    要:目的探讨表皮生长因子(EGF)在神经干细胞培养中的作用。方法取新生SD大鼠神经干细胞用无血清培养技术进行培养、传代和鉴定,在传代且培养基撤除EGF后观察细胞的变化,并进行神经元特异性烯醇化酶(NSE)、神经胶质酸性蛋白(GFAP)检测,以做细胞鉴定。结果成功培养出新生大鼠神经干细胞,并进行传代,培养的神经干细胞在培养基去除EGF后能分化为神经元细胞和神经胶质细胞,EGF反应性神经干细胞主要向神经胶质细胞分化。结论新生大鼠神经干细胞能在体外适宜条件下进行长期培养、传代,且具有多向分化潜能;EGF具有促进神经干细胞快速增殖、维持神经干细胞状态、抑制神经干细胞分化的功能;EGF反应性神经干细胞具有向星形胶质细胞分化的趋势。

Abstract:Objective To study the effect of epidermal growth factor for culturing neural stem cell.Methods The growth cycle and growth state for primary culture neural stem cells were observed carefully;after removing the epidermal growth factor from the culture media,the characteristics of differentiation of the neural stem cells were examined with immunofluorescence.Results The neural stem cells were incubated,and their development of cycle was investigated.The differentiated cells expressed NSE and GFAP,characteristic for neurons and glial cells.Conclusion The neural stem cells derived form newborn rat brains in this study are able to proliferate and multipitential for differentiation.Epidermal growth factor can inhibit the differentiation of the neural stem cells and accelerate the proliferation of neuron stem cell.EGF-responsive neural stem cells are prone to differentiate to astrocytes.
