摘    要:目的观察高脂饲料喂养的大鼠心脏组织中src抑制的C激酶底物(SSeCKS)的表达变化,探讨高脂血症对SSeCKS表达的影响。方法高脂饮食组(n=8)和正常饮食组(n=8)SD大鼠分别经高脂饲料和普通饲料喂养16周后,测血清总胆固醇及甘油三酯;用HE染色观察心脏病理学改变;用免疫组织化学法检测SSeCKS在心脏的表达;用免疫荧光双标法观察SSeCKS在心脏中的细胞定位。结果高脂饮食组总胆固醇和甘油三酯较正常饮食组明显升高(P〈0.05);病理学观察发现高脂饮食组形成高脂性心脏病变;免疫组织化学检测发现高脂饮食组中SSeCKS表达主要分布在心内膜、心间质;免疫荧光双标发现SSeCKS与心脏组织内皮细胞和α-平滑肌肌动蛋白阳性细胞部分共定位。结论高脂饲料喂养能使大鼠形成高脂血症,并引起心脏SSeCKS表达升高,SSeCKS可能参与了心脏结构重塑及细胞凋亡,从而促进动脉粥样硬化心脏病的发展。

Abstract:Objective To explore the effects of hyperlipidemia in high-fat diet rat on expression of src suppressed C kinase substrates(SSeCKS) in heart tissue.Methods SD rats fed with a high-fat diet were taken as high-fat diet group(n=8,high-fat diet group) and those fed with a normal diet as the control(n=8,normal diet group).Sixteen weeks later,serum lipid profiles including totoal cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL) and low density lipoprotein(LDL) were detected.Heart was stained with HE.SSeCKS expression was measured by immunohistochemistry and immunofluorescence.Results Compared with the control group,the model group showed elevated TC and TG(both P0.05).Further immunohistochemistry analysis revealed SSeCKS expression was significantly increased in heart in the model group whereas in the group little or no immuno-detectable SSeCKS expression was showed.Immunofluorescence detected SSeCKS colabled with endothelial cell marker CD31.Conclusion High-fat diet may lead to hyperlipidemia in rats,which may up-regulate SSeCKS expression in heart tissue.
