溴氯哌喹酮抑制角膜新生血管的实验研究

摘    要:Objective To evaluate the inhibitive effect of halofuginone on mouse corneal neovascularization.Methods Eyedrops of 500 μg/ml and 1000μ g/ml halofuginone were prepared with BSS.Mouse dry food containing 5mg halofuginone per kilogram was ready to feed mice.Uniformly sized hydron sustained pellets containing basic fibroblast growth factor(bFGF)were prepared and implanted into the C5781/6 mouse corneal tunnel.Mouse was administered with these eyedrops or fed with halofuginone food at normal manner.Control group was treated with BSS and fed routine diet.Corneal responses were monitored with slit-lamp biomicroscopy.The length and area of corneal neovascularization were measured through 10 days.The comparison between groups was done.Results The inflammation in cornea and anterior chamber were absent.The corneal neovascularization began to grow on day 3 from dilated blood vessel net of corneal limbus and reached the implanted pellet in stroma tunnel on day 5 to 7 with appearance of fan-shape.The neovascularization began to regress on day 10.The length and area of corneal neovascularization in food group were significant smaller than control group through observation period.These parameters in food group Were also smaller than eyedrop groups on day 3 and 7.The response of dose-dependent between two eyedrop agenm was not found.Conclusions Halofuginone could effectivemy inhibit the mouse corneal neovascularization.The activity of halofuginone was primarily due to inhibition of collagen al(I)synthesis and deposition,matrix metalloproteinsase(MMP-2)expression as well inhibition of smooth muscle and endothelial cell proliferation.
