温针灸对兔膝关节退变模型血清MMP-1和MMP-13水平的影响

目的观察温针灸治疗兔膝关节退变模型前后血清基质金属蛋白酶1、13（MMP-1,MMP-13）及软骨形态的变化,探讨温针灸治疗膝骨性关节炎的作用机理。方法 30只雄性兔随机分为模型组、温针灸组和西药组,每组10只,采用右后肢伸直位固定制作兔膝关节退变模型。采用双抗体夹心酶联免疫吸附法（ELISA）检测造模前、治疗前和治疗后兔血清中MMP-1,MMP-13水平,参考彭太平法观察膝关节组织形态学变化。结果治疗2周后,大体和光镜观察模型组膝关节软骨变薄,出现明显的关节软骨损伤,温针灸组可明显改善关节软骨组织学结构。各组治疗前较造模前血清MMP-1、MMP-13的浓度均显著升高（P〈0.05）;治疗两周后,与模型组比较,温针灸组MMP-1、MMP-13水平降低（P〈0.05）,但与西药组比较,差异无统计学意义（P〉0.05）。结论温针灸能治疗KOA的软骨损伤,可能与降低MMP-1和MMP-13表达有关。

Effects of Needle Warming Moxibustion on Serum MMP-1, MMP-13 Level in the Knee Osteoarthritis of Rabbits

Objective To investigate the influence of needle warming moxibustion on matrix metalloprotenases and histomorphology of the rabbit experimental knee osteoarthritis model, and to explore its relative mecha- nism. Methods 30 male rabbits were randomly divided into model, needle warming moxibustion and western medicine group, each group of 10 cases. The concentrations of MMP - 1, MMP - 13 in serum were detected by ELISA after two weeks treatment. Results The cartilage became thinning and appeared a significant articular injury. Needle warming moxibustion group can improve the histological structure of articular cartilage. MMP - 1 and MMP - 13 before treatment were increased significantly compared with normale （ P 〈 0.05 ）. Compared with model group,both MMP- 1 and MMP- 13 levels in warm acupuncture- moxibustion group were reduced considerablly （ P 〈 0.05 ）. No significant differences were found between western medication group and model group, western medication group and needle warming moxibustion group. Conclusion MMP - 1, MMP - 13 participated in the occurrence of KOA and needle warming moxibustion can treat KOA ar- ticular injury, which may related with the expression of MMP - 1 , MMP - 13.
