非洛地平逆转代谢综合征大鼠心肌纤维化的作用

摘    要:目的 探讨非洛地平逆转代谢综合征(MS)心肌纤维化的作用机制。方法 雄性Wistar大鼠33只随机分为对照组(12只)和代谢综合征组(21只),采用10%高果糖饮水8个月建立MS动物模型,随机将9只代谢综合征组大鼠给予非洛地平。并采用苦味酸天狼猩红染色法检测大鼠左室心肌胶原含量,RT-PCR法检测心肌白细胞介素-18(IL-18)mRNA表达水平。结果 与对照组相比,MS大鼠左室心肌组织胶原含量升高(P<0.01);IL-18 mRNA表达水平升高(P<0.01);应用非洛地平干预治疗后,上述异常均减轻。结论 非洛地平通过抑制IL-18表达在逆转MS心肌纤维化发生发展中起重要作用。

Abstract:Objective    To investigate the mechanism of reversal of cardiac fibrosis in fructose-fed rats (FFRs) by Felodipine. Methods    Thirty-three male Wistar rats were randomly divided into the control group (n=12) and the metabolic syndrome (MS) group (n=21). After the MS model was built by feeding with 10% fructose water for 8 months, 9 FFRs were perfused into the stomach with Felodipine once daily. Then, myocardium tissues were collected. Collagen content was detected by Sirius Red staining and real-time RT-PCR was to detect mRNA expression of interleukin-18(IL-18). Results    The collagen content of MS group was significantly higher than that of the control group (P<0.01).  mRNA expression of IL-18 was significantly higher in the MS group than in the control group (P<0.01). After six-week treatment of Felodipine, the collagen content and IL 18 mRNA expression were significantly lower than those in the MS group. Conclusion    Felodipine ameliorates cardiac fibrosis in MS via its suppressive effect on IL 18 expression.
