摘    要:【目的】 研究胰岛素增敏剂吡格列酮对小剂量地塞米松诱发大鼠出现的胰岛素抵抗的改善作用。【方法】大鼠皮下注射地塞米松(0.1 mg/kg,2次/d)的同时给予吡格列酮(20 mg/kg)灌胃,观察吡格列酮对胰岛素敏感性、葡萄糖耐量、血中甘油三酯含量及体重的影响。【结果】吡格列酮可明显提高小剂量地塞米松处理大鼠对外源性胰岛素的反应性,改善其异常的糖耐量,降低血甘油三酯水平,并使体重下降的程度减轻。【结论】吡格列酮可明显改善小剂量地塞米松处理大鼠的胰岛素抵抗。

Abstract:[ Objeclive] To investigate the effects of pioglitazone on insulin resistance induced by low dose dexamethasone in rats. [Methods] Rats were subcutaneously administered with low dose dexamethasone (0.1 mg/kg, twice daily) for 14 days and effect of pioglitazone at the dose of 20 mg/kg on glucose tolerance tests, insulin tolerance tests, serum triglyceride level, and body weight were observed. [ Regalts] Significant insulin resistance, impaired glucose tolerance, and mild weight loss were observed in low dose dexamethasone-treated rats. Pioglitazone treatment significantly decreased serum triglycefide level ( P 〈 0.05), and weight loss was inhibited ( P 〈 0.05) and insulin semifivity, impaired glucose tolerance was improved markedly ( P 〈 0.05). [ Conclusions] The data suggest that treatment with pioglitazone improves insulin sensitivity in low dose dexamethasone- treated rats. The insulin sensitizing effect may be associated with ameliorating lipid metabolism.
