摘    要:目的建立驱虫斑鸠菊高效液相色谱法指纹图谱(HPLC-FPs)分离的优化条件。方法采用HPLC法,用C18柱(150mm×4.6mm,5μm),流动相为乙腈-0.5%磷酸水溶液,检测波长为220nm,柱温为30℃,考察驱虫斑鸠菊水提取物在不同梯度洗脱条件下的分离情况。结果优选出的驱虫斑鸠菊HPLC-FPs分离条件,可使驱虫斑鸠菊水提物中各组分达到较佳分离,稳定性、精密度、重现性好。结论该色谱分离条件可用于建立驱虫斑鸠菊药材HPLC-FPs的分离条件。

Abstract:Objective To optimize the separation conditions of fingerprint for Vernonia anthelmintica(L.) Willd by high performance liquid chromatography and lay a foundation of studies on its fingerprint by high performance liquid chromatography. Methods A reversed-phase high performance liquid chromat-ographic method was adopted.The samples were extracted by aqueous solution and separated by a linear gradient elution on a C18 column(150 mm×4.6 mm, 5 μm),with the acetonitrile-0.5% phosphoric acid solution as mobile phase. The eluent was detected at UV 220 nm and the column was maintained at 30 ℃ .Du-ring the linear gradient elution program, the volume fraction of acetonitrile in mobile phase changed as follows:0-20 min,from 5% to13%;20-80 min,from 13% to 25%; the flow rate was 1 mL·min-1.Results The separation conditions described above achieved good results in the aspects of separation effect,stability,sensitivity,and reproducibility.Conclusion The optimum separation conditions of fingerprint would be useful for establishing the method of fingerprint for Vernonia anthelmintica(L.) Willd by high performance liquid chromatography.
