摘    要:目的测定野黄芩苷的血药浓度。方法RP HPLC法。采用VydacC8柱( 2 5 0mm×4 6mm ,5 μm) ,甲醇 乙腈 10mmol·L-1乙酸铵(体积比18∶9∶73,pH 2 5 )为流动相,流速为1 0mL·min-1,检测波长335nm ,柱温为30℃。结果Beagle犬静脉给予野黄芩苷后,野黄芩苷的血药质量浓度在0 0 5~12 5mg·L-1内与其峰面积有良好的线性关系(r =0 9986 )。结论该方法可用于野黄芩苷的临床前药物动力学研究。

Abstract:Objective To establish a method for determination of scutellarin in dog plasma by HPLC.Methods Vydac C 8 analytical column was used the mobile phase consisted of methanol,acetonitrile,10 mmol·L~ -1 ammonium acetate (V∶V∶V=18∶9∶73,pH 2.5).The flow rate was 1.0 mL·min~ -1 .The detector was operated at 335 nm.The column temperature was 30 ℃.Results The calibration curve was linear in the concentration range from 0.050~ 12.50 mg·L~ -1 (r= 0.997 5 ).The low limited quantification was 0.050 mg·L~ -1 .The inter-day and intra-day precision (RSD) of low,medium,high concentration (0.125,1.00,10.00 mg·L~ -1 ) of scutellarin in plasma was less than 7.9%.The accuracy (RE) was -1.0%, -2.8%, -1.7%, respectively.Conclusions The method is proved to be accurate,simple,and suitable for the preclinical pharmacokinetics scutellarin in dogs.
