| 摘 要： | 目的:研究黄连所含成分盐酸小檗碱的HPLC测定方法。方法:采用BDSHypersilC18色谱柱(4.6mm×250mm,5μm);流动相:乙腈-0.033mol/L磷酸二氢钾(含0.5%三乙胺,用磷酸调节pH至3.0)(30:70);检测波长:254nm;流速:1.0ml·min-1;柱温:室温;样品采用甲醇加热回流提取。结果:盐酸小檗碱在0.2336～1.7520μg范围内呈良好的线性关系,R2=0.9997(n=6),平均回收率99.17%,RSD=1.8%(n=5)。结论:该方法简便、准确、重现性好,可用于黄连药材中盐酸小檗碱含量的控制方法。

| Abstract: | Objectiue：To Establish method for HPLC determination of Berberine hydrochloride in Rhizoma Coptidis. Methods：BDS Hypersil C18 column （4.6mm×250mm,5μm）; mobile phase： Acetonitrile-0.033mol / L potassium dihydrogen phosphate （containing 0.5% triethylamine, pH adjusted to 3.0 with phosphoric acid） （30：70） ; detection wavelength： 254nm; flow： 1.0ml o min-1; column temperature： room temperature; sample heating reflux extraction using Methanol. Result： 0.2336 - 1.75201xg Berberine hydrochloride was well within the linear relationship, R2=0.9997 （n=6）, average recovery of 99.17%, RSD = 1.8%（n=5）.Conclusion： This method is simple, accurate, reproducible, and can be used for medicinal herbs in the Berberine hydrochloride content in Rhizoma Coptidis control methods. |
