清道夫受体对小鼠脂质代谢的影响

摘    要:目的观察清道夫受体AⅠ/Ⅱ(SR-AⅠ/Ⅱ)对小鼠脂质代谢的影响。方法采用SR-AⅠ/Ⅱ基因敲除(SR-AⅠ/Ⅱ-/-)小鼠及其同系对照(SR-AⅠ/Ⅱ+/+)小鼠,以高脂膳食(HFD)饲养12周后,采用酶法检测血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、载脂蛋白B(ApoB)等血脂水平,油红O染色法检测小鼠肝脏脂质沉积。结果SR-AⅠ/Ⅱ-/-组血清TG、TC、LDL-C、ApoB水平显著高于SR-AⅠ/Ⅱ+/+组(P<0.01),肝组织冰冻切片组织学分析显示,SR-AⅠ/Ⅱ-/-小鼠肝脏异常主要表现为肝细胞肿胀、大量脂肪沉积,且均较SR-AⅠ/Ⅱ+/+小鼠严重。结论SR-AⅠ/Ⅱ可能参与小鼠脂质代谢调节。

Effect of scavenger receptor class A type Ⅰ/ Ⅱ on lipid metabolism in mice
Abstract:Objective To observe the effects of scavenger receptor class A type Ⅰ/Ⅱ on lipid metabolism in mice. Methods TG, TC, HDL-C, LDL-C,and ApoB levels, were measured by enzymatic determination. Lipid accumulation in mice liver was analyzed by the lipid-specific Oil Red O-positive staining of frozen liver sections. Results The levels of serum TG, TC, LDL-C and ApoB of SR-AⅠ/Ⅱ knockout mice were more higher than those of wild type mice (P<0.01). The histological analyses revealed hepatocytes SR-AⅠ/Ⅱ deficient mice were distended and massive lipids SR-AⅠ/Ⅱ deficient mice accumulated in liver, and both SR-AⅠ/Ⅱ deficient mice were more severer than those of the wild type mice. Conclusion Scavenger receptor class A type Ⅰ/Ⅱ could play an important role in regulation of lipid metabolism in mice.
