摘    要:目的研究野生型和bikunin基因敲除小鼠卵丘-卵母细胞复合体显微和超微结构变化。方法用扫描电镜和光镜对诱导排卵后野生型和bikunin基因敲除小鼠卵丘-卵母细胞复合体结构上的变化进行观察。结果 (1)光镜显示,野生型小鼠中,卵丘细胞紧密围绕在卵母细胞周围;bikunin基因敲除小鼠中,卵丘细胞松散地分布在卵母细胞周围。(2)电镜显示,野生型小鼠可见大量的纤维网格状结构,卵丘细胞亦可见;bikunin基因敲除小鼠中,卵丘细胞之间未见有网状结构,卵丘细胞紧密连接少。(3)bikunin基因敲除小鼠卵丘-卵母细胞数明显低于野生型。结论 bikunin基因敲除小鼠卵丘细胞之间、卵丘细胞与卵母细胞之间结构发生变化,卵丘-卵母细胞数明显下降,该现象可能与卵丘-卵母细胞复合体的功能密切相关。

Abstract:Objective To study the ultrastructural changes of cumulus-oocyte complexes in wild-type and bikunin-null mice.Methods Female mice(wild-type and bikunin-null mice,aged 3~4 weeks) were instimulated to superovulation.The cumulus oocytes complexes were isolated and observed under light microscope and scanning electron microscope.Results(1) Light microscopy showed that the cumulus cells were poorly distributed around the occytes of bikunin-null mice,and intensely organized around the occytes of wild-type mice.(2) In the wild-type mice,the cumulus cells viewed under SEM showed a highly branched filamentous substructure;but in the bikunin-null mice,intimate connection among the cumulus cells was rare and the highly branched filamentous substructure was absent;(3)The number of the cumulus-oocyte complexes in the bikunin-null mice was significantly lower than in the wild-type mice.Conclusion Morphological changes of cumulus cells and cumulus oocytes in bikunin-null mice are closely related to the function of the cumulus oocyte complexes.
