弓形虫DNA疫苗在免疫鼠体内的分布情况研究

摘    要:目的观察弓形虫疫苗pcDNA3.1-P30-P22免疫小鼠后在小鼠体内的分布情况。方法雌性BALB/c系小鼠,随机分成3组,每组12只,第1组为pcDNA3.1-P30-P22免疫组,第2组为空质粒pcDNA3.1对照组,第3组为磷酸盐缓冲液(PBS)对照组;免疫组经肌内注射免疫,于免疫后4周和8周,分别取小鼠的注射部位肌肉、血液、肝脏及脾脏抽提组织DNA,并以此为模板进行多聚酶链反应(PCR)扩增,扩增产物经琼脂糖凝胶电泳分析。结果免疫后4周,免疫组小鼠的上述器官均扩增出P30-P22基因片断,对照组均未扩增出相应片断,与预期结果相符;免疫后8周,仅免疫鼠血液扩增出上述特异性片断。结论弓形虫DNA疫苗pcDNA3.1-P30-P22免疫小鼠后,重组质粒能被多个组织摄取,但在不同组织中存留的时间不同。

Abstract:Objective To explore the distribution of the DNA vaccine pcDNA3.1-P30-P22 of toxoplasma gondii in immunized mice.Methods The plasmids were injected into the BALB/c mice via muscles,then the muscle,blood,liver and spleen were sampled after 4 and 8 weeks of immunization to extract tissue DNA as templates,and then to amplify the gene fragments by means of polymerase chain reaction(PCR)and to identify the amplifiction products by agarose gel electrophoresis.Results After 4 weeks of immunization,P30-P22 gene fragments were amplified out of those tissues of the vaccinated mice,which were in accordance with the expected results.The control group mice could not have any fragment amplified.But the specific gene fragments were only amplified from the blood of the vaccinated group after 8 weeks of immunization.Conclusion The combinant plasmids ditribute in different sites of the mice immunized by DNA vaccines of Toxoplasma gondii,but the lasting time is different.
