川芎化学成分及其抑制子宫平滑肌收缩作用

摘    要:目的研究川芎Ligusticum striatum DC.的化学成分及其抑制子宫平滑肌收缩作用。方法川芎水煎液正丁醇部位采用硅胶、ODS、Sephadex LH-20、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物结构。采用缩宫素致大鼠离体子宫平滑肌痉挛模型考察化合物对子宫平滑肌收缩活动的影响。结果从中分离得到8个化合物,分别鉴定为环(L-苯丙氨酸-L-亮氨酸)(1)、环(L-苯丙氨酸-L-缬氨酸)(2)、环(L-苯丙氨酸-L-异亮氨酸)(3)、尿嘧啶(4)、胸腺嘧啶(5)、6-羟甲基-3-羟基吡啶(6)、烟酸(7)、腺嘌呤(8)。与空白组比较,化合物1~3均在高剂量时可以抑制子宫收缩张力和收缩活力(P<0.05),中、低剂量组对子宫平滑肌收缩无明显作用。结论化合物1~3、6~7首次从该植物中分离鉴定。化合物1~3对离体子宫平滑肌收缩活动有抑制作用。

Abstract:AIM To study the chemical constituents from Ligusticum striatum DC.and their inhibitory effects on uterine smooth muscle contraction.METHODS The n?butanol fraction of aqueous extract from L.striatum was isolated and purified by silica,ODS,Sephadex LH?20,and reversed phase semi?preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The rat uterine smooth muscle model was used to determine the effects of the compounds on the contractile activity of uterine smooth muscle,which was induced by oxytocin.RESULTS Eight compounds were isolated and identified as cyclo?(L?Phe?L?Leu)(1),cyclo?(L?Phe?L?Val)(2),cyclo?(L?Phe?L?Ile)(3),uracil(4),thymine(5),6?hydroxymethyl?3?pyridinol(6),nicotinic acid(7),adenine(8).Compared with the blank group,compounds 1?3 could inhibit uterine contractile tension and contractile activity at the high dose(P<0.05),while the medium and low dose groups had no significant effect on uterine smooth muscle contractile.CONCLUSION Compounds 1?3,6?7 are isolated from this plant for the first time.Compounds 1?3 inhibit contractile activities of isolated uterine smooth muscle.
