摘    要:目的建立HPLC法同时测定不同产地山茱萸果核中没食子酸、莫诺苷、5-羟甲基糠醛、原儿茶酸的含有量。方法山茱萸果核60%乙醇提取液的分析采用Thermo Hypersil GOLD AQ C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.2%甲酸水溶液,梯度洗脱;体积流量1.0 mL/min;检测波长270 nm;柱温30℃。结果没食子酸、莫诺苷、5-羟甲基糠醛、原儿茶酸分别在7.67~57.51(r=0.999 9)、1.79~13.40(r=0.999 8)、0.78~5.85(r=0.999 8)、5.06~37.98(r=0.999 9)μg/mL范围内线性关系良好,平均加样回收率分别为99.61%、103.74%、98.31%、99.56%,RSD分别为3.00%、2.25%、2.42%、2.09%。结论该方法准确稳定,重复性好,可用于山茱萸果核的质量控制。

Abstract:AIM To establish an HPLC method for the simultaneous content determination of gallic acid,morroniside,5-hydroxymethylfurfural,protocatechuic acid in the fruit core of Cornus officinalis Sieb. et Zucc. from different growing areas by HPLC.METHODS The analysis of 60% ethanol extract of the fruit core of C. officinalis was performed on a 30 ℃ thermostatic Thermo Hypersil GOLD AQ C18 column(4.6 mm×250 mm, 5 μm), with the mobile phase comprising of acetonitrile-0.2% aqueous formic acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 270 nm.RESULTS Gallic acid,morroniside, 5-hydroxymethylfurfural,protocatechuic acid showed good linear relationships within the ranges of 7.67-57.51(r=0.999 9),1.79-13.40(r=0.999 8),0.78-5.85(r=0.999 8),5.06-37.98(r=0.999 9) μg/mL,whose average recoveries were 99.61%,103.74%,98.31%,99.56% with the RSDs of 3.00%,2.25%,2.42%,2.09%,respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of the fruit core of C. officinalis.
