肺腺癌患者血浆microRNA-10b表达及临床意义

目的 探讨肺腺癌患者血浆microRNA-10b（miR-10b）与临床特征的关系及其作为肺腺癌肿瘤标志物的可行性。方法 46例肺腺癌患者（肺腺癌组）与42例体检健康者（对照组）,采用实时荧光定量PCR检测2组血浆miR-10b水平,分析miR-10b与肺腺癌临床特征间关系。绘制ROC曲线,评估miR-10b的诊断效能。结果 肺腺癌组血浆miR-10b水平（1.632（0.836,2.313））较对照组（0.497（0.221,0.766））上调3.284倍（P〈0.01）;miR-10b表达在淋巴结转移上差异有统计学意义（P〈0.05）,在肿瘤原发灶情况、TNM分期上差异无统计学意义（P〉0.05）;miR-10b为1.212时,诊断肺腺癌的敏感性为64.3%,特异性为82.4%,AUC为0.765。结论 肺腺癌患者血浆miR-10b表达明显上调,miR-10b有望成为肺腺癌的早期诊断标志物。

Expressions of plasma microRNA-10b in patients with lung adenocarcinoma and its clinical significance

Objective To investigate the relationship of microRNA-10b （miR-10b） level in plasma with clinicopathological features and its feasibility of being one of tumor markers for lung adenocarcinoma. Methods The expressions of miR-10b in plasma were assayed by real-time quantitative PCR in 46 patients with lung adenocarcinoma and 42 normal controls. The relationship between miR-10b expression and clinical characteristics was analyzed. ROC was drawn to evaluate the diagnostic value of miR-10b to the diagnosis of lung adenocarcinoma. Results The expression of plasma miR-10b of lung adenocarcinoma （1. 632 （0. 836, 2. 313）） was 3. 284 times as high level as normal controls （0. 497 （0. 221, 0. 766）） （P〈0.01）. The expression of miR-10b showed a significant difference in lymph node metastasis （P〈0.05） and no significant difference in the primary focus and TNM staging （P〉0.05）. When miR-10b was 1. 212, the sensitivity was 64.3%, the specificity was 82.4%, and AUC was 0. 765. Conclusion The plasma miR-10b level is up-regulated in patients with lung adenocarcinoma, which suggests miR-10b might be used as a potential tumor marker for the early diagnosis of lung adenocarcinoma.
