MK-801并胚胎脊髓移植促进大鼠脊髓损伤后功能恢复的实验研究

摘    要:目的研究胚胎脊髓移植同时应用NMDA受体拮抗剂MK-801观察其是否能够促进半切洞脊髓损伤运动功能恢复.方法将成年大鼠分为三组,A组单纯脊髓半切洞损伤组.B组脊髓半切洞损伤+胚胎脊髓移植组,C组脊髓半切洞损伤+胚胎脊髓移植+MK-801组.手术后应用联合行为评分(CBS)、感觉诱发电位(SEP)、运动诱发电位(MEP)检查.结果三组CBS得分A组>B组>C组,SEP和MEP潜峰时A组>B组>C组,统计学分析有显著差异性.结论通过分析CBS、SEP、MEP结果表明胚胎脊髓移植和NMDA受体拮抗剂MK-801能够促进脊髓损伤后功能恢复.

Abstract:Objective: To study Whether acute treatment with NMDA antagonists MK-801 would improve motor function in rats with hemisected spinal cord and received transplants of fetal spinal cord into the hemisection site. Methods: Adult rats were divided into three groups. Group A received lumbar spinal hemisections only. Group B received lumbar spinal hemisections and fetal (embryonic day 14) spinal cord grafts and Group C received lumbar hemisections and fetal (embryonic day 14) spinal cord grafts and treated with MK-801 at the time of hemisections and graft. After operation combined behavioral score (CBS), somatoserisory evoked potential (SEP) and motpr evoked potential (MEP) were examined. Results : The CBS score sequence among group were found in three groups and they were A>B>C (p<0.05). The peak latencies of early waves in SEP and MEP sequence among Group A, Group B and Group C were A>B>C (P<0.05). Conclusion: CBS, SEP and MEP change assessments suggested that grafts of fetal spinal cord tissue with NMDA receptor antagonists MK-801 could promote the recovery of neural function of adult rat spinal cord.
