摘    要:目的采用次全结扎巴马猪冠状动脉法建立心肌缺血模型。方法对10只巴马小型猪术前行冠状动脉CTA(CCTA)及心脏MR(CMR)检查。开胸后次全结扎冠状动脉左前降支建立模型,并于术后2、12、24、48、72h及1、2、3、4周复查CTCA及CMR,测心功能指标,包括左心室舒张末期容积(LVEDV)、收缩末期容积(LVESV)及射血分数(LVEF)。术后4周处死实验动物行病理检查。结果7只巴马小型猪顺利完成手术,建模成功率70.00%。建模后CTCA示结扎处狭窄程度约80%~100%;与术前比较,术后24h后不同时间点LVEF下降(P0.05)、LVESV增加(P0.05),术后2周后不同时间点LVEDV增加(P0.05)。CMR检测梗死节段与病理结果的一致性较好(Kappa=0.888)。结论次全结扎冠状动脉左前降支可建立良好的心肌缺血动物模型。

Abstract:Objective To establish myocardial ischemia model in Bama mini-pig by partial ligation of the coronary artery. Methods Ten Bama mini-pigs were detected with CT coronary angiography (CCTA) and cardiac magnetic resonance (CMR) before operation as the baseline. Then myocardial ischemia model was established through partial ligating the left anterior descending coronary artery. CCTA and CMR were performed 2 h, 12 h, 24 h, 48 h, 72 h, 1 week, 2 weeks, 3 weeks and 4 weeks after modeling, respectively. The cardiac function was measured with CMR, including left ventricular end-systolic volume (LVESV), left ventricular end-diastolic volume (LVEDV) and left ventricular ejection fraction (LVEF). Finally, pathologic histology was performed. Results Myocardial ischemia model was successfully established in 7 of 10 mini-pigs, and the success rate was 70.00%. After modeling, CTCA showed the degree of coronary stenosis was about 80%-100%. Compared the cardiac function of all periods with the baseline results, statistical significances were observed as follows:LVEF declined 24 h after modeling (P < 0.05), LVESV increased 24 h after modeling (P < 0.05), while LVEDV increased 2 weeks after modeling (P < 0.05). Areas of delayed-enhanced in CMR were consistent with pathologic findings (Kappa=0.888). Conclusion Myocardial ischemia model can be well established by partial ligation of the left anterior descending coronary artery in Bama mini-pig.
