黏液瘤病毒对裸鼠子宫内膜癌移植瘤的抑瘤作用及机制

摘    要:目的 探讨黏液瘤病毒对裸鼠皮下移植子宫内膜癌的抑瘤作用及其可能机制.方法 采用皮下注射子宫内膜癌HEC-IB细胞的方法建立裸鼠子宫内膜癌模型.瘤内注射黏液瘤病毒后,观察肿瘤生长变化,并用免疫组化方法检测肿瘤组织中PI3K、Akt、GSK表达水平及活性的变化.同时通过瘤内注射灭活病毒和生理盐水作为对照组.结果 黏液瘤病毒对正常裸鼠不存在毒性及致病能力.黏液瘤处理组肿瘤大小较对照组缩小,差异具有统计学意义.经黏液瘤病毒处理的肿瘤组织中PI3K、Akt的表达水平较对照组显著降低.结论 黏液瘤病毒对裸鼠子宫内膜癌移植瘤具有抑瘤作用,其可能机制是通过调节PI3K/AKT(磷脂酰肌醇3激酶/丝氨酸苏氨酸激酶)信号通路调控细胞凋亡.

Abstract:Objective Explored the effects of Myxoma virus in endometrial cancer of Nude Mouse animal model in vivo.Methods CD-1 Nude mice were randomly divided into three groups with 8 mice eachgroup and established mice models of endometrial carcinoma,and group M was administrated by1 × 107PFU MV(10 μl)QOD,the other 2 group (group D and group B) was administrated by 10μl dead virus and normalsaline,respectively.The time of tumorigenesis,tumorformationrates and the tumor volumes were observed,and theP13K,Akt expression changes in nude mice models of endometrialcarcinoma were assessed by immunohistochemical method.Results The endometrial cancer in all nude mice were established successfully.And tumor growth in group M were significantly decreased than group D and group B after injection,some tumor were lessened,the expression of PI3k,Akt were decreased in group M.Conclusion Administration of MV into mice models of endometrial carcinoma resulted in the reduction of tumor volumes by suppressing tumor angiogenesis and the PI3K/Akt path-way which is related to the tumor cell proliferation.
