川芎-香附提取物对原发性痛经模型动物的作用

摘    要:目的探讨3种不同提取方式提取川芎-香附药对所得到的提取物对动物原发性痛经模型的药理作用,为川芎-香附药对治疗痛经的新药开发提供依据。方法采用缩宫素致大鼠离体子宫剧烈收缩模型,观察各提取物对大鼠离体子宫活动的影响;采用小鼠原发性痛经模型以扭体次数和扭体潜伏期为指标,观察提取物的镇痛效果。结果 3种提取物能不同程度抑制大鼠离体子宫自发活动,对缩宫素引起的离体子宫剧烈收缩的抑制活性依次为超临界CO2萃取物>醇提水沉液>水提醇沉液。3种提取物均能不同程度减少缩宫素致痛经模型小鼠的扭体次数,延长扭体潜伏期。结论超临界CO2萃取川芎-香附药对(分子蒸馏纯化)所得到的提取物对大鼠离体子宫剧烈收缩模型及小鼠原发性痛经模型具有明显抑制及镇痛作用。

Abstract:Objective To study the pharmacological action of three different extracts from Chuanxiong Rhizoma and Cyperi Rhizoma on dysmenorrhea models in murine. Methods The isolated rat uterus model was used to study the effect of the extracts on the contractile activity; the oxytocin-induced dysmenorrhea models in mice were used to study the analgesic effects of the extracts according to the number and latent period of twist body. Results The extracts had certain inhibition to the spontaneous activity of isolated uterus, and the inhibition rates were as the following order: supercritical CO2 fluid extraction> alcohol extract-water supernate > water extract-alcohol supernate. The extracts could decrease the number of twist body and prolong the latent period of twist in oxytocin-induced mice dysmenorrheal model. Conclusions Supercritical CO2 fluid extract from Chuanxiong Rhizoma and Cyperi Rhizoma shows remarkable inhibition effect on the contractile activity of isolated rat uterus and oxytocin-induced dysmenorrheal model in mice.
