In vitro and in vivo Effects of Praziquantel on Cysts of Echinococcus granulosus

Both in vitro and in vivo praziquantel damages the germinal layer of secondary cysts in mice infected with protoscoleces obtained form sheep naturally infected with metacestodes of Echinococcus granulosus.Praziquantel-induced damage of the germinal layer was characterized by degeneration,lysis and vesiculation of the matrix in the tegument of the cysts,In addition,emer-gence of lysosome-like structures,lipid droplets,and increase in the number of mitochondria ac-companied by swelling and degeneration in the perinuclear cytoplasm,as well as extensive lysis andloss of the innermost layer bordering the cysts cavity were also noted. However, when infected mice were treated with praziquantel in vivo,the efficacy of praziquantel as measured by mean cyst weight was unreliable as a decrease in mean cyst weight was apparent in only a few experiments.Therefore,the cysts of E.granulosus appear less susceptible to praziquantel.
