不同灸法对免疫抑制兔脾脏指数及细胞因子的影响

目的：观察不同灸法对环磷酰胺所致免疫抑制兔脾脏指数及细胞因子 IFN-γ、IL-2及 IL-4的影响。方法将大耳白兔50只随机分为空白组、模型组、隔药饼灸组、艾灸组和假药饼灸组5组。除空白组外，其余4组均腹腔注射环磷酰胺60 mg/kg ，造成免疫抑制模型。3个治疗组分别给予隔药饼灸、艾灸、假药饼灸治疗，隔日灸，共灸10次。治疗结束次日动物麻醉，取动物脾脏计算其脾脏指数，采用 ELISA 法测定匀浆上清液中 IFN-γ、IL-2、IL-4含量。结果环磷酰胺造模后，脾脏指数与空白组比较显著升高，差异具有显著的统计学意义（P＜0.01），3个治疗组脾脏指数与模型组比较差异均有显著统计学意义（P＜0.01），与空白组比较差异无统计学意义（P＞0.05）。与空白组比较，艾灸组 IFN-γ、IL-4明显降低，模型组、假药饼组IL-4明显降低，差异具有统计学意义（P＜0.05）。与模型组比较，艾灸组 IFN-γ明显降低，隔药饼灸组IL-4显著升高，差异具有统计学意义（P＜0.05）。结论不同灸法对免疫抑制兔脾脏指数均有调节作用；隔药饼灸可使因环磷酰胺所致下降的IL-4显著提升。

Influence of Different Moxibustion on Spleen Index and Cytokines in Immunosuppressive Model Rabbits

Objective To observe the inhibitory effect of different moxibustion methods on rabbit spleen index, the contents of IFN-γ, IL-2 and IL-4 in cyclophosphamide induced immunosuppression model rabbits. Methods The 50 rabbits were randomly divided into five groups, namely black group, model group, herb-cake-separated moxibustion group, moxibustion group, s ham-cake-separated moxibustion group. In addition to the black group, the other four groups were injected with cyclophosphamide 60 mg/kg to make immunosuppressive model. Three treatment groups were respectively given herb-cake-separated moxibustion, moxibustion, sham-cake-separated moxibustion treatment every other day, for 10 times. The next day after finished moxibustion, the animals were anesthetized, the spleen index was detected, the IFN -γ, IL -2, IL -4 in supernatant of spleen homogenate were determined by ELISA. Results Compared with the black group, the spleen index of model group was significantly higher, it was statistically significant (P<0.01). The spleen index in the three treatment group was statistically significant comparing with the model group (P<0.01), while the diffrence between the treatment groups and blank group was not statistically significant (P>0.05). Compared with the blank group, the IFN-γ and IL-4 in the moxibustion group were lower significantly, the IL-4 in model group and sham-cake-separated group was reduced, the differences were statistically significant (P<0.05). Compared with the model group, the content of IFN-γ was decreased in moxibustion group, and the content of IL-4 increased in herb-cake-separated moxibustion group was increased, the differences were statistically significant (P<0.05). Conclusion Different moxibustion shows a regulating effect on spleen index in immunosuppressive rabbits. The herb -cake -separated moxibustion could significantly increase the decline content of IL-4 induced by cyclophosphamide.
