姜黄素对瘦素刺激HSC增殖及JAK2-STAT3信号通路影响的研究

摘    要:目的:观察姜黄素对瘦素刺激肝星状细胞(HSC)增殖及对JAK2-STAT3信号通路的影响。方法:SD大鼠36只灌胃姜黄素煎剂7 d,制备各药物含药血清,用噻唑蓝(MTT)比色法检测药物血清对100 pg·L~(-1)的瘦素刺激HSC-T6增殖的影响;应用蛋白印迹试验检测姜黄素散对100 pg·L~(-1)瘦素刺激HSC-T6中JAK2、STAT3蛋白的表达水平。结果:姜黄素药物血清有抑制HSC-T6的作用(P0.05或P0.01));姜黄素作用6 h后,JAK2、STAT3蛋白的表达水平开始出现降低,且JAK2降低较为明显,24 h后STAT3降低最为明显。结论:姜黄素药物血清具有抑制瘦素刺激HSC-T6增殖的作用;而JAK2-STAT3通路被阻断,JAK2-STAT3蛋白表达的降低可能是其发生的主要机制之一。

The Research into Influence of Cur on Leptin stimulating the HSC Proliferation and JAK2 and STAT3 Information pathway
Abstract:Objective:To observe the influence of cur on leptin stimulating the HSC proliferation and JAK2 and STAT3 information pathway.Methods:SD rats were given Cur decoction for gavage for 7 days to prepare the drugs containing medicine serum,then the influence of serum containing medicine on HSC-T6 proliferation stimulated by 100ng/ml of leptin were detected by using MTT colorimetric method;And the influence of Cur Powder on JAK2 and STAT3 protein expression levels in HSC-T6 stimulated by 100ng/ml of leptin were measured by using protein imprinting test.Results:Serum containing Cur has function of inhibiting HSCT6(P<0.05 or P<0.01));After cur is used for 6 hours,the expression of JAK2 and STAT3 protein levels started to reduce,and JAK2 reduce more significantly;After 24 hours,STAT3 reduced the most obviously.Conclusion:Serum containing Cur has the function of inhibiting HSC-T6 proliferation stimulated by leptin.JAK2 and STAT3 pathway is blocked,JAK2-lower STAT3 protein expression may be one of the main mechanisms.
