摘    要:目的建立更昔洛韦自微乳化微球中更昔洛韦含量测定的高效液相色谱方法。方法采用自微乳化技术和锐孔凝固渗法,将更昔洛韦制成自微乳化微球。色谱柱选用zorbax SB-C18(2.1×100nm,3.5nm)。流动相为磷酸二氢钠缓冲液-甲醇95%:5%,流速为1.0ml/min,波长252nm,柱温为25℃。结果更昔洛韦标准曲线方程在0.75—100.00ng/ml浓度范围内,线性关系良好;相对回收率均在98.28%~101.60%范围内,绝对回收率大于98.31%;日内和日间精密度RSD均小于3.3%。结论本方法具有操作简便、测定速度迅速、试剂价格便宜等优点,适用于更昔洛韦自微乳化微球中更昔洛韦含量的测定。

Determination of Ganciciovir in Self-microemulsified Microspheres by HPLC

Abstract:Objective To develop a high performance liquid chromatography method for the determination of ganciclovir in self-mieroemulsified microspheres. Methods Using ganciclovir to prepare self-microemulsion microspheres by micro-emulsion technology and orifice solidification osmosis, column using Zorbax SB-C18(2.1 × 100 nm,3.5 nm). The mobile phase consisted of sodium dihydrogen phosphate buffer and methanol 95% :5% ,the flow rate was 1.0 ml/min,wavelength 252 nm,column temperature was 25%. Results Detection of ganciclovir standard curve equation was 0.75 - 100.00μg/ml concentration range, good linear relationship between the relative; recovery rate was 98.28% -101.60% range, with absolute recovery more than 98.31% ;the intraday and interday precision RSD were less than 3.3%. Conclusion This method had the advantages of simple operation, fast determination speed, and cheap reagent price, which was suitable for determination of ganciclovir in self-microemulsifted microspheres.
