HPLC-ELSD同时测定柿叶中齐墩果酸及熊果酸含量

目的：建立测定柿叶中齐墩果酸和熊果酸含量的高效液相色谱-蒸发光散射检测法（HPLC-ELSD）.方法：以甲醇为提取溶剂,色谱柱为Waters SunFire^TM C18（250 mm×4.6 mm,5μm）,流动相为乙腈-甲醇-1.0％醋酸铵溶液（55∶25∶20）,流速为1.0 mL·min-1,柱温为25℃.蒸发光散射检测器漂移管温度为90℃,气体压力为0.17 MPa.结果：齐墩果酸的量在0.132 5～1.656 μg呈良好线性关系（r=0.999 8）,回收率为98.86％;熊果酸的量在0.370 4 ～4.630 μg呈良好线性关系（r=0.998 6）,回收率为98.88％.结论：所建立的方法简单准确、重现性好,可用于柿叶的质量控制.

Simultaneous Determination of Oleanolic Acid and Ursolic Acid in Kaki Folium by HPLC-ELSD

Objective： To establish a high performance liquid chromatography-evaporative light scattering detector （HPLC-ELSD）method for the determination of oleanolic acid and ursolic acid in Kaki folium. Methods： The samples were extracted by methanol and the determination was performed on Waters SunFireTM C18 column （250 mm×4.6 mm,5μm） with mobile phase consisted of acetonitrile-methanol - 1.0% ammonium acetate（55：25：20, V/V）at the flow rate of 1.0 mL.min-1 and column temperature of 25 ℃, The detection was performed on an evaporative light scattering detector with the drift tube temperature of 90 ℃ and a gas pressure of 0. 17 MPa. Results： Oleanolic acid showed a good linear relationship at the range of 0. 132 5 - 1. 656 μg（r =0. 999 8）. The average recovery was 98.86%. Ursolic acid showed a good linear relationship at the range of 0. 370 4 -4. 630 μg（r =0. 998 6）. The average recovery was 98.88%. Conclusion： The established method is simple and accurate, with good reproducibility, and can be used for quality control of Kaki folium.
