二期梅毒患者外周血淋巴细胞凋亡及其Fas、FasL表达的研究

摘    要:目的:检测二期梅毒患者外周血淋巴细胞(PBLC)凋亡及其Fas、FasL的表达,探讨二期梅毒患者细胞免疫功能异常与淋巴细胞凋亡的关系。方法:采用流式细胞仪检测33例二期梅毒患者和30例正常人PBLC细胞凋亡及其Fas、FasL的表达。结果:梅毒患者CD3+、CD4+及CD16,56+细胞凋亡率显著高于对照组(P<0.01),CD8+细胞凋亡率两组比较差异无显著性(P>0.05)。梅毒患者CD3+、CD4+及CD16,56+细胞Fas与FasL表达均明显高于对照组(P<0.01),CD8+细胞Fas与FasL表达两组比较差异无显著性(P>0.05)。梅毒患者CD3+、CD4+及CD16,56+细胞凋亡率分别与其Fas、FasL表达呈显著正相关(r=0.685,P<0.01;r=0.712,P<0.01;r=0.705,P<0.01;r=0.694,P<0.01;r=0.716,P<0.01;r=0.727,P<0.01)。结论:二期梅毒患者细胞免疫功能异常可能与淋巴细胞凋亡过度有关,而淋巴细胞凋亡过度与Fas、FasL表达异常有关。

Abstract:Objective. To investigate the correlation between apoptosis of peripheral blood lymphocytes(PBLCs) and impaired immunity assessing the apoptosis and expression of Fas and FasL in PBLCs of patients with secondary syphilis. Methods.Flow cytometry (FCM) was used to detect PBLC apoptosis as well as Fas and FasL expression in 33 patients with secondary syphilis and 30 normal controls. Results.The apoptosis levels were significantly higher in CD3^+, CD4^+and CD16,56^+ cells in patients than those in controls ( P〈 0. 01), and was not different in the CD8^+ cells between patients and controls (P〉0.05). The Fas and FasL expressions on CD3^+ ,CD4^+ and CD16,56^+ cells were significantly higher in patients than that in controls(P〈0. 01). No significant difference was found in either Fas and FasL on CD8^+ (P〉0. 05). In patients group, the apoptosis levels of CD3^+ , CD4^+ and CD16,56^+ cells had positive correlations with the Fas and FasL expression, respectively(r=0. 685, P〈0. 01 ; r=0. 712, P〈0. 01 ; r= 0. 705, P〈0. 01 ; r= 0. 694, P〈0. 01 ;r=0. 716,P〈0. 01 ;r=0. 727, P〈0. 01). Conclusions: Dysfunction of cellular immunity may be due to the increased PBLC apoptosis in patients' with secondary syphilis, which is associated with the abnormal expression of Fas and FasL on PBLCs.
